Mary Campbell was accepted at Kansas State University before season finale, but she decided to go on a trip to discover her identity without the influence of all the alternative versions she saw in the portal with Dean and the Impala. Mary could well decide to go and study at university if she and John settle down somewhere.
On their travels, they have the freedom to choose their own destiny as Dean encouraged them to do. John was gifted Dean's own hunting journal and Mary the colt capable of taking down the yellow-eyed demon but did they end up needing to use these gifts? They could have used both to defend themselves and stay safe, or they could have continued their hunting career.
In Supernatural, it's less John and Mary finding the yellow-eyed demon and more the other way around. Yellow eyes is able to sneak into their family home and infect the infant Sam with Demon blood. Equipped with the vast knowledge that Dean would have collected in his time hunting and the weapon to destroy this awful demon, John and Mary have their best chance at success in defending their family from the evil demon.
Rowena makes a cameo in season 1 when she provides Ada with a crystal capable of killing the Akrida Queen. Rowena trades this weapon to Ada in exchange for a plant that holds a demon captive and mentions that she needs to find out where her son is from the demon.
